Background: Age-Related Molecular Changes

By the time most people reach their fifties, several measurable biochemical shifts are already underway. Peroxisomal activity — the enzymatic machinery responsible for synthesizing a class of specialized lipids called plasmalogens — begins to decline. [10] Mitochondrial efficiency changes with age. [5] Published studies have observed that PlsEtn levels in certain brain regions tend to be lower in older individuals compared to younger cohorts, and that this difference is more pronounced in individuals with certain neurological conditions. Because the peroxisome is the primary organelle responsible for synthesizing the plasmalogen backbone, [10] declining peroxisomal enzyme activity with age may affect the body’s endogenous plasmalogen synthesis capacity. The science

Magnesium is a cofactor for ATP hydrolysis, for peroxisomal DHAP-acyltransferase activity (a step in plasmalogen biosynthesis), [10] and for mitochondrial membrane stabilization. [17] Population data suggest that dietary magnesium intake is below recommended levels in a substantial proportion of older adults, a gap attributable in part to reduced dietary intake and increased urinary excretion with age. [21]

In a preclinical study, elevating brain magnesium via magnesium L-threonate was associated with increased synaptic density and improved learning measures in rodent models. [18] Observational studies in human populations have found associations between higher serum magnesium and measures of cognitive health [19] and inflammatory markers. [20] These findings are associative and do not establish causation or indicate that magnesium supplementation prevents any disease.

[23]

Vitamin K2 supports the gamma-carboxylation of osteocalcin (a bone matrix protein) [25] and matrix Gla-protein (MGP), which is involved in inhibiting calcium deposition in vascular tissue. CoQ10 facilitates electron transfer from Complexes I and II to Complex III of the respiratory chain, supporting the proton gradient that drives ATP synthesis. [31] Research suggests that when CoQ10 availability is limited, mitochondrial ROS generation may increase as a byproduct of impaired electron flow. [32]

Endogenous CoQ10 levels are observed to decline with age in several tissues. [29] Statin medications, which inhibit the mevalonate pathway, also reduce CoQ10 synthesis as a secondary effect. [29] Ubiquinol, the reduced form, is directly bioavailable without requiring enzymatic conversion in the liver, which may make it preferable for older individuals. [30]
